About the Michaelis-Menten Constant (Km)

The Michaelis-Menten constant (Km) is a fundamental parameter in enzyme kinetics. It represents the substrate concentration at which the reaction rate is half of its maximum value (Vmax). A lower Km indicates a higher affinity of the enzyme for its substrate.

The formula used in this calculator is:

Km = (k₋₁ + k₂) / k₁

Where:

  • k₁ is the rate constant for substrate binding
  • k₋₁ is the rate constant for substrate unbinding
  • k₂ is the rate constant for product formation
